I tested it after finding a save and doing the quest. The staff does benefit from Twin Souls. However, it seems that Sanguine Rose has the Summoner perk already built in because the range of the Dremora is massive. I reckon that to get the most from the staff it could be that you get to Twin Souls by going up the Necromancy side of the skill tree.
